Poon Pik Wan joined the HKRep in 1986. She furthered her studies in acting and Putonghua at the Central Academy of Drama in 1995 and trained under the tuition from performing arts tutors of Mainland China, Germany and the United States.
She appeared in over 70 productions, including Cat On a Hot Tin Roof, A Midsummer Night's Dream, Twelfth Night, Marriage of Figaro, Peer Gynt, the musical Cabaret, Tales of the Walled City, Ricky My Love, The Story of Qu Yuan, Shall We Love, Red Sky, De Ling and Empress Dowager Ci Xi, Small Ripples in a Dead Pool, Forever and Ever, A Matter of 4x6, A Dream Like a Dream, Love In a Fallen City, Tomorrow Is Another Day, The Eccentricities of a Nightingale, Haunted, Haunted Little Star, Departure 00:00, Sweet & Sour Hong Kong, A Small Family Business, Peach Blossom Fan, The Importance of Being Earnest, February 14 and Vassa Zheleznova, (Rep 2 Series) Angel in the Box and Xi Nai on Rock.

Poon was the director's assistant of The School For Scandals, Romance in the Teahouse, A Midsummer Night's Dream and Secret of Resurrection and was the assistant director of www.com.

Poon was nominated the Best Actress (Tragedy / Drama) at the 2004 HK Drama Awards for her work of Alma Winemiller in The Eccentricities of a Nightingale and she was awarded Best Supporting Actress (Tragedy/Drama) for her works of Peach Blossom Fan in 2005. She was nominated Best Actress (Tragedy/Drama) at 15th HK Drama Awards in 2006 for Vassa Zheleznova.

At 16 th HK Drama Awards In 2007, she was nominated Best Actress (Tragedy / Drama) for her work of the Ophthalmologistˇ¦s Wife in Blindness (World Premiere).
